Neurobiology of Human Sexuality Human sexuality is a topic that has been taboo for many centuries. The orgasm, for example, is one aspect of sexuality that has been overlooked. In particular, the female orgasm is an important function to address because it's a subject that has been socially steeped in silence and shame, and is directly connected to female health in general. Not many people are willing to admit, that orgasms actually benefit a woman physically and psychologically in many ways (i.e. easing menstrual cramps to alleviate stress). Instead, they allow themselves to have orgasms. Not only that, but they can have a variety of different types of orgasms at the same time. There is a combination of things that come in effect when it comes to an orgasm in the brain. Neurotransmitters, and neuro-peptides for example, are heavily involved in the sexual response. To reach an orgasm, the central nervous system sends orders to the heart, so that it pumps faster, sending blood to oxygenate the increase of blood flow in muscles involved in sexual activity. Nitric oxide, serotonin, dopamine, epinephrine, and norepinephrine, are just a few of the neurotransmitters and neuro-peptides involved in sexual activity. Nitric oxide (NO) plays a critical role in both male and female sexuality. In penile erections, NO stimulates the release of guanylate cyclase, which, in turn, converts GTP to cGMP and produces relaxation of smooth muscles and increased blood flow into the penis. This information is used in the popular drug sildenafil (ViagraTM) that inhibits the metabolism of cGMP to prolong the effects of the erection. Serotonin's role in sexual function is that of constriction of smooth muscles in the genitals, and peripheral nerve function. Epinephrine appears to be involved in maintaining the penis in a flaccid state. This increases the rate and force of the muscle's contractions during sexual activity. Conversely, in women, epinephrine has been shown to increase vaginal pulse amplitude. Norepinephrine is another neurotransmitter that mediates chemical communication in the sympathetic nervous system, a branch of the autonomic nervous system. Like other neurotransmitters, it is released at synaptic nerve endings to transmit the signal from a nerve cell to other cells. Levels in the brain of neropinephrine vary according to sexual arousal. They increase significantly with arousal and sexual activity in men as well as in women.

Hormone levels are also involved in sexual activity. In males, testosterone levels remain generally above the threshold required for sexual interest and activity. Thus increases testosterone above this threshold are believed to have additional influences on sexual interest or behavior. Interestingly, estrogen(the "female hormone") seems to have little impact on sexual desire on either males or females. Estrogen deficient women, however, can cause a decrease genital lubrication caused by the thinning of the vaginal epithelium. All factors can impair both the physiological and psychological aspects of sexual arousal.

Other hormonal influences in sexual function include oxytocin, cortisol, pheromones, and prolactin. There is some evidence showing that oxytocin (produced by dopamine) levels increase during sexual arousal and orgasm in both men and women. Also the combination of oxytocin and female hormones like estrogen in women, encourage an emotional attachment with a partner. In men, the bonding effect is muted, due to the male's higher testosterone levels. As a result, men form emotional ties slower, and are more likely to regard initial sexual contact as just sex. Studies of men and women with elevated levels of prolactin report decreased sexual interest, arousal, orgasm as well as mood disturbances such as anxiety and depression. Dysfunction in remaining erect has been described in men with both abnormally low and abnormally high prolactin levels. In women, high prolactin has been associated with infertility and decreased sexual activity. Animal studies also suggest that prolactin has an inhibitory influence on both male and female sexual behavior, although short-term prolactin elevations may play a role in some elements of sexual behavior in male rats. Clearly, this molecule plays many roles in humans but the exact role is uncertain. It is known that prolactin shares a close connection with dopamine in the brain, particularly in the tubero-infundibular tract, which runs along the base of the hypothalamus and releases dopamine into the portal veins of the pituitary gland. Dopamine acts to inhibit the release of prolactin. This, in turn, affects dopamine release. Blockade of dopamine receptors in the tuberoinfundibular tract releases prolactin from the tonic inhibitor control of dopamine, allowing prolactin levels to rise. This self-regulation is critical to prolactin homeostasis, since any disturbance in the connection between the hypothalamus and the pituitary may lead to hyperprolactinemia (high prolactin levels).

Knowing what happens in the brain during sexual arousal and orgasm can help to speculate why so many men and women have problems during sexual activity. There are various factors for both men and women that can cause sexual dysfunction such as depression(caused by low level of serotonin), everyday stress, etc. Understanding the sexual differences in both men and women, can also help differentiate male and female behaviors. But there is still on-going research being done on these issues because not much in known about how these chemicals work together in the brain to affect sexual behavior. There are speculations for example, that the olfactory sensors in men are highly sensitive to the "smell" of estrogen and in fact, cause quick arousal. But this kind of triggering in the "female" brain is less receptive. Perhaps this is why women have to "think" about having an orgasm in order to feel one. Whether women control their orgasm or not, is still questionable. But it is obvious that the levels and ways that these chemicals work together in the brain are different for men and women.

Blogger 'not welcome' here: Canadian mayors tell Roosh V

The mayors of several major Canadian cities are adding their voice to the growing backlash against a controversial American blogger who’s behind a series of men’s meetups planned for this weekend.

The so-called “pick-up artist” Daryush Valizadeh, known online as “Roosh V,” also runs the website Return of Kings, which is described as a forum for “heterosexual, masculine men.”

An international Return of Kings meetup day is scheduled to take place in 43 countries around the globe, including 10 Canadian cities, on Saturday.

On Tuesday, Ottawa Mayor Jim Watson took to Twitter to publically tell Return of Kings supporters that their “pro-rape, misogynistic, homophobic garbage is not welcome in Ottawa.”

Watson is also asking venues in the city to deny renting space to the group to hold the meetup.

A number of other Canadian mayors quickly followed suit and decried the meetings.

Effects of Long-term Treatment of Butea superba on Sperm Motility and Concentration

Abstract

Butea superba has been used to enhance sexual performance in men with unknown long-term effects on spermatozoa. This study aimed to investigate effects of chronic treatment of B. superba on sperm motility and concentration in rats and mice in correlation with testicular damage. Adult male rats and mice were orally administered distilled water or B. superba alcoholic extract (0.01, 0.1 or 1.0 mg/kg BW/day) for 6 months. At the end of the treatment the animals were sacrificed and the spermatozoa recovered from the distal cauda epididymis were analyzed for sperm concentration and motility in vitro. The testes were processed for histological examination. The results show that long-term treatment with B. superba extract significantly increased the sperm concentration and delayed the decreased motility with time. None of signs of sperm anomalies and testicular damages were observed. This suggests that chronic use of B. superba increases the number of sperm, prolongs sperm motility in vitro while produces no changes on sperm morphology. Therefore, chronic use of B. superba alcoholic extract may be useful in fertilization.

Introduction

Butea superba, known as "Kwaao Khruea Daeng" in Thai, has been used for physical and mental strength and for preventing age-related health problems. Thai traditional medicine uses B. superba as a rejuvenating agent and to increase sexual performance in men (Suntara, 1931). The plant preparation appears to improve an erectile function in patients with erectile dysfunction (ED) without any apparent toxicities (Cherdshewasart and Nimsakul, 2003). Fertilization in vivo requires adequate numbers of spermatozoa to be ejaculated with normal morphology and motility (Adamopoulos et al., 1996). Since spermatogenesis involves a complex process of cellular development, impairment in any of the development stages may lead to a reduction in the fertility. Long-term utilization of any sexual enhancers may affect the amount of and/or characteristics of spermatozoa resulting in altered fertilization capability. Therefore, this study aimed to investigate effects of chronic treatment of B. superba alcoholic extract on sperm motility and concentration in correlation to testicular damage in rats and mice.

Materials and Methods

Extract preparation

B. superba tuber roots were collected from Phrae, Thailand, and identified by Associate Professor Yuttana Smitasiri, School of Science, Mae Fah Luang University, Thailand. The roots were washed and air-dried at room temperature. The root cores were separated and sliced into small and thin pieces. Then they were dried in a hot air oven at 50o C and grinded into fine powder. The powder was extracted 3 times with ethanol at 1:4 ratio w/v and stirred overnight at room temperature. The ethanolic extract was evaporated under reduced pressure at 45o C using a rotary evaporator. The extract was dissolved in distilled water before used. Animals Male Sprague-Dawley rats (8 wks) and ICR mice (8 wks) were obtained from the National Laboratory Animal Center, Mahidol University, Nakornpathom, Thailand. They were housed under a 12:12-hour light-dark cycle and maintained at 24 + 1o C.

Extract treatment

Groups of 6 animals were randomly divided into four groups: one control and three different treatment groups. The control rats and mice were orally administered distilled water daily for 6 months. The treatment groups received alcoholic extract of B. superba at the doses of 0.1, 1.0 or 10.0 mg/kg BW daily for 6 months. Preparation of culture drop Motility test was performed in the tissue culture medium 199 (TCM199) modified with Earle's salt with L-glutamine (Life Technologies, USA) and supplemented with 3 mg/ml bovine serum albumin (Sigma, USA). Sperm motility evaluation was performed in a 100-µl droplet under 10 ml paraffin oil in a 60-mm petri dish (Nunclon, Denmark) and equilibrated overnight in high humidified atmosphere at 5% carbon dioxide (CO2) in air before further analysis.

Sperm motility assay

Sperm motility was assessed by the method of Bavister and Andrews (Bavister and Andrews, 1988). The distal cauda epididymis was dissected out and placed in 2 ml of equilibrated medium in a 35-mm tissue culture dish. A needle was employed to release the spermatozoa from the cauda epididymis into the medium. The spermatozoa were allowed to swim out into the medium for 3 min in the CO2 incubator at 37o C. Sperm concentrations were then determined. The medium was added to the culture drop for sperm motility assay providing a final concentration of 2 x 106 spermatozoa/ml. The tissue culture dish was placed under an inverted microscope (Nikon, Japan) and an observation was performed at a magnification of 20X within 1 min. The motility of sperm was observed at various time intervals up to 6 hrs.

Sperm count and morphology

Testicular histopathology The testis was dissected out, cut into small slices, and fixed in 10% formaldehyde buffer for 24 hrs. The tissues were washed free of 10% formaldehyde and stored in 70% alcohol until being embedded. The tissues were dehydrated in alcohol series and embedded in paraffin. Tissue sections of 5 µm thickness were prepared and placed on glass slides. The sections were stained with hematoxylin and eosin and mounted in mounting medium. The slides were examined under the light microscope. For each testis several cross sections composing of 20-50 tubule sections were examined for signs of interstitial edema, seminiferous tubule degeneration, and congestion.

Statistical analysis

Statistical analysis was performed using one way analysis of variance (ANOVA). If a significance was found, differences among individual group means were tested by the least significant difference (LSD) test. Values were considered statistically significant at P<0.05. The cauda epididymis was cut and weighed. A cell suspension was prepared by macerating the cauda in 1.0 ml of 0.85% saline. The cell suspension was kept for 24 hrs at 4o C. The suspension was then filtered through a double gauze layer and an aliquot of the sample was used for sperm count in a Neubauer hemocytometer. An aliquot of the epididymal sperm suspension was smeared and stained with hematoxylin and eosin and then examined under a light microscope (CH-2, Olympus, Japan) at magnification of 100X. The head and tail abnormalities (200 sperms per animal) were recorded.

Results

Effect of B. superba on sperm motility Motility of the sperms from the control rats and mice at time zero were about 90% (Figures 1 and 2). The movement gradually declined with time and reduced to 70% at 6 hrs in both species. Long-term treatment of the animals with B. superba alcoholic extracts at the doses of 0.01, 0.1 or 1.0 mg/kg/day significantly delayed the reduction of the sperm motility compared to the control group. The motility of sperms from the treated animals remained over 75% at the sixth hour after being investigated. However, at the time zero, the extract-treated groups revealed no difference in motility compared to the control group.

Discussion and Conclusion

Sperm motility has been considered as one of the most important predictors of fertility. Several reports have demonstrated the correlation of motion parameters with fertilization rates (Check et al., 1990; Liu et al., 1991). Adenosine triphosphate (ATP) is the main energy source of the sperm motility. A reduced ATP production and/or reduced ATP levels via ATPase hydrolysis result in an insufficient energy and poor sperm motility. This study shows that B. superba prolonged the sperm motility via unknown mechanisms. It is possible that B. superba may increase the ATP production and/or utilization. In addition, it cannot be ruled out that the plant may act on the nitric oxide (NO)-cyclic adenosine monophosphate (cAMP) pathway. A number of studies have reported involvements of the NO and cAMP in the sperm motility. A nitric oxide synthase (NOS) inhibitor (NG -nitro-L-arginine methyl ester) (Lewis et al., 1996) and a NO scavenger (methylene blue) (Donnelly et al., 1996) were reported to inhibit movement of human spermatozoa. Sildenafil, an inhibitor of phosphodiesterase V, increases the velocity and amplitude in human spermatozoa (Lefievre et al., 2000). Sildenafil also dose-dependently increases cAMP production in the spermatozoa. Human spermatozoa subjected to capacitating conditions increase their endogenous NO synthesis and their intracellular cAMP content. The cAMP synthesis is increased by NO releasing compounds and decreased by NOS inhibitors (Belen Herrero et al., 2000). These studies suggest an involvement of the NO and cAMP in the movement of the spermatozoa. B. superba has been reported to inhibit phosphodiesterase activity in vitro (Roengsumran et al., 2000). Therefore, it is possible that B. superba may also prolong the sperm motility by increasing the cAMP production. However, further studies are required to confirm the proposed mechanisms of action of B. superba.

An assessment of a sperm motility loss over time is normally used as a quality control of an in vitro culture medium used in clinical analysis of Assisted Reproductive Technology (ART). Some laboratories employ sperm survival or stress tests to enhance the predictability of the in vitro fertilization rate (Alvarez et al., 1996; Coccia et al., 1997). Therefore, the motility results from this study are valid for further use in clinical study design of B. superba. The prolonged sperm motility in the culture medium by B. superba extract indicates that B. superba may be useful in infertile men. The number of spermatozoa was increased by B. superba extract. This may be caused by β-sitosterol component in B. superba. The enzymes in the testis convert β-sitosterol to pregnenolone, an important substrate of testosterone synthesis (Subbiah & Kursis, 1975). The testosterone activates the release of gonadotropin-releasing hormone (GnRH) from hypothalamus. The follicle-stimulating hormone (FSH) and luteinizing hormone (LH) released by the GnRH induce the spermatogenesis and growth of spermatozoa.

Pro-rape' US pick-up artist posts personal details and pictures of female journalists online in revenge for negative coverage

A controversial 'pro-rape pick-up artist' is posting the personal details of journalists who have criticised him online.

Daryush Valizadeh - also known as Roosh V - is infamous for arguing that raping women should be legal on private property.

Labelled 'Operation Bullhorn', Roosh has asked his online supporters to 'adopt' a journalist and post their details on his forum. They have been instructed to gather photos, Facebook profiles and have even been told to save addresses for possible future use.

One forum user said the backlash was 'because women are scared that they won't be able to get a free lunch anymore by virtue of having a vagina.'

The backlash follows criticism of international meetups which included eight UK cities, including Manchester, London, Leeds, and Glasgow.

The meet-ups, set to take place today, were cancelled after Roosh claimed he feared for the safety of his supporters. ----

In recent years, the popularity of ‘vaginal rejuvenation’ surgeries, which includes vaginal tightening (vaginoplasty) and labial reduction or reconstruction (labiaplasty), have seen a significant increase. This is unsurprising, since mainstream media and culture constantly drills the notion into women that they can always look better, right down to their genitals. Moreover, several notable celebrities, such as Sharon Osbourne and TOWIE’s Gemma Collins, have openly talked about undergoing such procedures to achieve what has been dubbed the ‘designer vagina’.

According to a study by the International Society of Sexual Medicine in 2011, 87% of women who underwent vaginal rejuvenation surgery did so for purely cosmetic reasons. However, it is important to point out that this type of surgery is not just about cosmetics. There is no such thing as a ‘perfect vagina’, as labia comes in all different sizes and shapes and as such, medical professional carrying out these surgeries should be cautious when promising to give women a beautiful-looking vagina.

Cosmetic motives aside, there are a number of functional and medical reasons as to why women may want to undergo vaginal rejuvenation surgery. Giving birth or ageing can result in a lax vagina, making a woman feel self-conscious, or causing physical problems like incontinence and lack of sexual gratification. An enlarged or distorted labia can also create a genuine physical problem for many women, with discomfort occurring during sex, exercise or simply through catching on clothes.

In these cases, where the genitals are causing a woman significant problems in her day-to-day life, labial and vaginal reconstructive surgery is often recommended. Women are fed unrealistic representations of what they should look like on a daily basis and it is for this reason that any good gynaecologist will discourage women from undergoing vaginal reconstructive procedures based on superficial reasons alone.

A top doctor is warning against a bizarre trend that suggests ground-up wasp nests will help tighten the vagina.

While some natural ingredients are commonly used and endorsed for home remedies, Canadian gynecologist Jen Gunter is warning against one in particular.

Some online retailers, including on Etsy, have been selling oak galls - nests which house wasp eggs - which, they say, should be ground into a paste for 'vaginal rejuvenation'.

Oak galls are formed when a gall wasp lays eggs in a tree's leaf buds and the larva will then develop while inside the gall.

The substance, retailers claim, will restore the uterine wall after childbirth, heal an episiotomy cut and also clean out the vagina.

But Dr Gunter has spoken out on her blog, insisting that it is not recommended and could lead to a slew of side effects - including painful sex, a lack of healthy bacteria and an increased risk of contracting HIV.

Dr Gunter slammed the practice calling it 'dangerous' on her blog.

She said: 'This product follows the same dangerous pathway of other 'traditional' vaginal practices.

'Drying the vaginal mucosa increases the risk of abrasions during sex (not good) and destroys the protective mucous layer (not good).

'It could also wreak havoc with the good bacteria. In addition to causing pain during sex it can increase the risk of HIV transmission. This is a dangerous practice with real potential to harm.'

Health website the Female Renewal Solution claims that oak gall can help prevent cervical cancer and is 'all you need' to make the vagina 'tighter instantly and overnight.'

Etsy retailer HeritageHealthShop hawked the product by citing South East Asia medicine claims that the galls could improve sex lives and be used on cuts.

The page, which has since sold out of the galls, suggested grinding the balls into a paste to be applied to episiotomy cuts, which is a surgical cut in the muscular area between the vagina and the anus.

It used to be routinely done before delivery to enlarge the vaginal opening to prevent the vagina from tearing during childbirth.

HeritageHealthShop does warn that the paste will hurt but said it was due to the 'galls' powerful astringent'.

Dr Gunter said: 'Here's a pro-tip, if something burns when you apply it to the vagina it is generally bad for the vagina.'

This isn't the first time Dr Gunter has cautioned against products claiming to clean, dry or tighten the vagina.

Last year she warned women not to use herbal 'womb detox' products because they were risking health problems including toxic shock syndrome.

The Herbal Womb Detox Pearls are still being sold worldwide by Florida-based firm Embrace Pangaea.

The site claims the products can help with conditions including endometriosis, ovarian cysts, thrush and fibroids.

Women are directed to use the bags of perfumed herbs three at a time, for 72 hours.

The pearls claim to 'cleanse the womb and return it to a balance[d] state' by flushing out toxins.

The company claims that by tightening the womb, the vaginal canal will shrink and 'can result in heightened sexual pleasure.'

The website warns virgins, pregnant women or those breast-feeding against using the product.

Dr Gunter said the 'pearls' were not only ineffective and inserting anything into the vagina for too long was 'dangerous and smelly.'

Describing the vagina as like a 'self-cleaning oven,' she explained how inserting herbs is likely to interfere with its natural balance, damaging the good bacteria, or lactobacilli and irritating the lining, increasing the risk of getting an infection.

Zimbabwe: Age of Consent to Go Up to 18 - Mnangagwa

VICE PRESIDENT Emmerson Mnangagwa says government was in the process of introducing a law that would see the statutory age of consent to sex by girls raised from 16 to 18 years.

He was responding to calls by Senators on Tuesday that the minimum age one could be allowed to consent to sex should be aligned to a 2016 Constitutional Court ruling which outlawed the marriage of or among minors below 18.

While the ruling was followed with euphoric victory among child and women's rights activists, some felt it remained hollow as girls as young as 16 could still indulge in sex and even conceive children for as long as they did not proceed to get married before 18.

"We have a landmark ruling in this country which states that nobody should be married or be married off when they are below the age of 18," Senator representing people with disabilities, Anna Shiri, had said earlier.

She was contributing to a motion which called on the government to ratify and incorporate into its gender laws, the SADC Model Law on Eradicating Child Marriage.

Zimbabwe, currently, is in the process of enacting a law which seeks to operationalise the outlawing of child marriages.

"At the same time we have this Bill," Shiri said, "while we have part of the Act which says there is an age of consent to sex which is 16 years of age.

"As a result, there is some contradiction where early marriage is 18 years yet the consent to sexual activity is 16 years. We need to align these laws."

Mnangagwa, who was speaking as the country's justice minister, said government was considering raising the age of consent to 18.

"I may also say that the other two points you have raised relating to the issue of consent between juveniles or children, anybody who is below 18 is regarded as a child," Mnangagwa said.

"We had a Committee to deal with that and we have arrived at a possible solution which will come to Parliament on the issue of consent between an adult and juvenile or between a juvenile and a juvenile.

"Those issues we have debated and we believe that we should bring up also the age of consent to the age of 18. Of course, this is subject to debate when it comes to Parliament."

Mnangagwa said his ministry was in the process of aligning all the marriage laws and the progressive provisions of the SADC Model law he said will be incorporated in the comprehensive proposed Marriages Bill.

The proposed Bill will be able to amend the Marriages Act and the Customary Marriages Act and all other laws that are outdated in relation to marriages.
